TEM image of epon section of a freeze-substituted mouse retina illustrating the unusual distribution of heterochromatin in rod receptor nuclei where the compact heterochromatin is centrally located. Within the heterochromatin, two regions were identified, and inner 'smooth' mass (smo), and an outer 'granular compartment (gra), each with a distinct optical diffraction pattern (see inserts).

Discs were punched from freshly dissected mouse retina, cryoprotectant added, the samples cryo-fixed using high pressure freezing using a Leica EMPACT2, freeze-substituted in acetone containing OsO4, and embedded in Epon. Sections were stained with uranyl acetate and lead citrate, and observed with a Tecnai F20 TEM, and tilt series recorded with a 2k CCD camera.
