Tbx20 is a transcription factor that is required for heart development. Its global deletion in mouse models results in severe cardiac malformations and early embryonic death. In order to elucidate the specific role of Tbx20 in the endocardium of the developing heart, we have generated a conditional knockout using Cre-lox recombination. Endocardial deletion of Tbx20 with Tie2-Cre disrupts outflow tract formation, resulting in a double-outlet right ventricle and lethality around E14. Microarray and ChIP analysis reveal differentially regulated transcriptional targets within endocardium. Our results implicatendicate that endocardial Tbx20 as a mediator of endocardial es crosstalk with adjacent myocardium. We consider a number of novel mechanisms by which this signaling may occur, such as Bmp-Smad pathways, modulation of extracellular matrix components, and non-canonical Wnt signaling
