Intracellular X-rhodamine actin polymerized into filaments at the very leading edge of the lamellipodium, resulting in the narrow red band along the tip of the lamelipodium. Lamellipodial F-actin was labeled with Alexa488-phalloidin using glutaraldehyde fixation subsequently to the X-rhodamine actin incubation. \nThe micrograph visualizes that the actin monomer incorporation into the lamellipodial actin filament structure is restricted to the front of the lamellipodium. This enables a directed extension of the branched actin network towards the plasmamembrane, which pushes the membrane forward thereby creating cell protrusion.\n\nWide field epifluorescence image taken on using a 100X 1.49 NA lens, Semrock DA\/FI\/TR\/Cy5-4x4M-B Sedat filter set, and an Orca II with 500 ms exposure."},"ITEMTYPE":{"onto_name":"recorded image","onto_id":"FBbi:00000265"},"CELLULARCOMPONENT":{"onto_name":"actin cytoskeleton","onto_id":"GO:0015629"},"RELATIONTOINTACTCELL":{"onto_name":"dispersed cells in vitro","onto_id":"FBbi:00000611"},"SOURCEOFCONTRAST":{"onto_name":"distribution of a specific protein","onto_id":"FBbi:00000597"},"MOLECULARFUNCTION":{"onto_name":"actin binding","onto_id":"GO:0003779"},"TERMSANDCONDITIONS":{"free_text":"public_domain"},"IMAGINGMODE":[{"onto_name":"widefield illumination","onto_id":"FBbi:00000277"},{"onto_name":"fluorescence microscopy","onto_id":"FBbi:00000246"}],"DIMENSION":[{"Space":{"Image_size":500,"Pixel_size":{"unit":"microns","value":0.064},"axis":"X"}},{"Space":{"Image_size":500,"Pixel_size":{"unit":"microns","value":0.064},"axis":"Y"}},{"Wavelength":{"unit":"nanometers","value":"488"}},{"Wavelength":{"unit":"nanometers","value":"561"}}],"NCBIORGANISMALCLASSIFICATION":{"onto_name":"Mus musculus","onto_id":"NCBITaxon:10090"},"PREPARATION":[{"onto_name":"glutaraldehyde fixed tissue","onto_id":"FBbi:00000011"},{"onto_name":"saponin permeabilized","onto_id":"FBbi:00000264"}],"VISUALIZATIONMETHODS":[{"onto_name":"Alexa Fluor 488","onto_id":"FBbi:00000440"},{"onto_name":"X-Rhodamine","onto_id":"FBbi:00000584"},{"onto_name":"phalloidin","onto_id":"FBbi:00000100"}],"CELLTYPE":[{"onto_name":"fibroblast","onto_id":"CL:0000057"},{"onto_name":"primary cell line cell","onto_id":"CL:0000001"}]}},"Citation":{"Title":"Ingo Thieverssen, Clare M.
